Difenhydramine
Has a H1 receptor blocker derived from etanolamine that acts by causing a competitive antagonism with liberated histamine in certain pathologic conditions but does not interfere with histamine synthesis and storing.

Its well absorbed after intramuscular administration and intravenous administrations improves a rapid action. Half life and distribution volume are increased by rising dose in almost all species, while plasmatic protein binding and total depuration do not vary.

Distribution volume is wide in almost all species end placental distribution is wide.

Intramuscular route is very effective and very safe, because side effects are rarely seen, and this is very important when intravenous administration is considered, because if it administered by this route rapidly, may cause hypotension or inhibit respiration.
It is biotransfomed in liver and to certain extent in lungs and kidneys.
It is indicated in all kind of allergies, such as: urticaria, asthma, stomatitis and some types of edema, as well as problems in which there is cell destruction and toxin liberation. Also in case of gangrenous mastitis, placenta retention, emphysema.
Because of it mucosarinic effect, it can be used in organophosphorate intoxication therapy.

Side effects may include sedation or excitation, gastrointestinal disorders, parasympaticlytic action, local anesthesia, allergies and theratogenies. At recommended dose and without combination with other drugs that increase potency it is relatively non-toxic.
It is probable to have an anti-arrhythmic effect.

It is now known that histamine diverse actions on the body are performed by combination with two kind of receptors. H1 through which have effect on smooth muscle en bronchi and intestine and on allergic reactions. And on H2 receptors through which have effect on gastric secretion, heart, uterus. Other histamine actions, as those performed on blood vessel muscles are performed by simultaneous effect on H1 and H2 receptors.

HISTAFIN® (difenhydramine) has a blocker of H1 receptors and is derived from etanolamine that acts by causing a competitive antagonism with liberated histamine in certain pathologic conditions but does not interfere with histamine synthesis and storing, and therefore histamine liberation is not reduced, but avoid that it acts on cells by competing with it in specific receptors.
